I've stared into the sun so many times I should be blind
but every single day I see I reckon with the time
I find the birds in hiding and I catch the birds in flight
they're everything we try to be, they settle for "alright"
how easily we sway because our human's flesh and bone
it leads us to believe we have been made to be alone  
tucks everything we feel into a the box that disappears
considers not the Spirit, faithful always through the years
let everywhere I need to be forgive where I will go
and leave behind the people I will never really know

I remember us
We talked. We laughed
We were happy to have each other
We have trusted one another
And I never thought
This might end.

I remember how
We spent so much time together
We talked about everything
Being open books to one another
And I never could imagine
This might end.

I remember those long nights
After having said "sleep well"
We were still talking about this and that
Even though both very tired the next day
We always did it again and again
And I never believed
This might end.

I remember being so happy with you
Laughing so much that it hurt
Trusting you, being there for you
Always wanting to keep that forever
-
Years and years. And at first
Our friendship grew stronger.
But having reached the highest point
The time and space became our enemies.
I never expected
This to have an end.

I know deep inside you understand
But you stopped chasing those good memories
Still it was hard for me to let them go
...
Anyway I just wanted to let you know
All my secrets and thoughts
All my losses and pains
I wanted to share with you
What was in my heart, in my veins
But you lost the belief in us
And we couldn't keep the trust

And I never again got to know
About how you feel deep inside
What you actually think in your mind
How you feel about losing your best friend
Because now our friendship seemed to end

And I always blamed the circumstances
But I knew that you changed a lot
So maybe if you didn't let go off
We could still make it through everything
Just how we used to do it once before -
When I never could imagine
This might end.
The friendship is unfortunately fading and I don't feel that I can save it anymore. Unfortunately. We picked to go different ways.

Are you still there?
Can you hear me?
I can almost feel you
skirting around the outside of my awareness
subsiding in my periphery
I still want you
  but not for my self
I still hear you
  but not speaking in prose to me
I still feel your fire
  but only as an afterthought
Days and months have passed
Still some small part of you remains
Will it be weeks and months
until your presence is gone?
Do I even want that..
Want you absent
Permanently
Unseen
Unfelt
Unheard
A ghost

It was the end of our days,
The closure of olden ways,
A real vintage train,
Souvenir of a passing parade,
A blast from the past,
We cheered as it rolled on, at last,
Shall we see those days again?
The revival of steam trains,
No, it was all part of change,
It was the end of our days,
The closure of olden ways.......

Preventing contamination,
A constant challenge in cell culture.
Contamination not only affects,
The culture in question and,
Costs time and money,
But also endangers the reproducibility of results.

No cell culture problem,
Is as universal as that of culture loss
Due to contamination.

Generally, contamination may be separated,
Into categories of microbial,
And eukaryotic contamination.

Examples of microbial contamination include:
Bacteria (including Mycoplasma),
Fungi and yeast;
Eukaryotic contamination includes:
Cross-contamination with other cell lines.

Bacteria, yeast and fungi,
The three more common types of contamination,
But luckily these forms are often detectable,
Under the microscope and,
By visual cues,
Like colour or turbidity changes in the medium.

Mycoplasma is a small genus of bacteria,
That lack a cell wall and for this reason,
They remain unaffected by common antibiotics.
They are also difficult to detect,
With standard microscopes,
Due to their size, about 0.1 μm in diameter,
And the fact that they often attach to host cells.


To prevent contamination,
Use 70% ethanol for disinfecting,
Equipment & surfaces,
Related to cell culture.
Sterile filter the media first,
Before bringing to the lab.

Fetal Bovine Serum,
A potential source of contamination,
Contains mycoplasma.
Filter it at 0.1 μm, or,
Gamma irradiate it.
Aseptic technique,
Necessary.

The laboratory workers be the last,
But not the least source of contamination.
Teach them the ideal laboratory practices,
To ensure asepticity in a laboratory.
